چکیده

The use of electrospinning technology has enabled wound dressings to be developed in an exceptionally advanced way. Biopolymer-made fibrous mats have been extensively studied for tissue engineering due to their similar structure to the extracellular matrix. Using scaffolds incorporating drug delivery represents an increasing interest in promoting tissue repair through controllable drug delivery. It has been reported that deferoxamine (DFO) improves wound healing by increasing the stability of hypoxia-inducible factor-1α (HIF-1α). In this study, a composite of Silk/Chitosan/PVA fibrous mat was prepared by electrospinning. polymer blend containing ciprofloxacin were formed into cross-linked nanofiber mats with three-dimensional properties. Afterward, with the aid of polydopamine (PDA) as a linker, deferoxamine (DFO), was immobilized on the surface of the as-prepared PVA /Silk /Chitosan fibrous membrane. Chemical, mechanical and in-vitro evaluations as well as the vivo study in rat model were clarified the characteristic of the mats. A comparison between pristine and modified PVA /Silk /Chitosan fibrous mat revealed that the modified version showed bactericidal effect and the angiogenic properties with the accelerated wound healing capability while possessed significant tensile properties through the introduction of ciprofloxacin and DFO and the presence of the silk in mats. The results of the presented study, suggesting the DFO modified PVA /Silk /Chitosan fibrous could be served as an effective dressing for treating diabetic ulcers.
